cyclin-dependent kinases (cdks) 4 , 6 enzymes have been shown promote cell division , multiplication in both normal , cancer cells. many cancer cells have shown abnormalities increase activity of cdk, leading inactivation of tumor suppressor genes.


when used in combination other drugs such alk or mek inhibitor, ribociclib has been shown have synergistic effect, resulting in improved responses. again, result of crosstalk between signaling pathways. blocking 1 pathway in cancer tumorigenesis can result in tumor compensation , tumor compensates blocked signaling pathway utilizing other pathways survive. blocking several pathways @ once, thought tumor less able compensate, , greater anti-tumor response observed. utilizing ribociclib in combination other agents has been shown reduce development of resistance these agents. in other words, cancer’s development of drug resistance can mitigated addition of ribociclib therapeutic regime.
