the production of particular digestive exoenzyme bacterial cell can assessed using plate assays. bacteria streaked across agar, , left incubate. release of enzyme surroundings of cell cause breakdown of macromolecule on plate. if reaction not occur, means bacteria not create exoenzyme capable of interacting surroundings. if reaction occur, becomes clear bacteria possess exoenzyme, , macromolecule hydrolyzed determines identity.


amylase

amylase breaks down carbohydrates mono- , disaccharides, starch agar must used assay. once bacteria streaked on agar, plate flooded iodine. since iodine binds starch not digested byproducts, clear area appear amylase reaction has occurred. bacillus subtilis bacterium results in positive assay shown in picture.


lipase

lipase assays done using lipid agar spirit blue dye. if bacteria has lipase, clear streak form in agar, , dye fill gap, creating dark blue halo around cleared area. staphylococcus epidermis results in positive lipase assay.
